Manual microplate-based bead cleanup is time consuming and prone to variability and errors, highlighting the need for automated solutions to enable efficient, reproducible high-throughput workflows. Bead-based cleanup steps are a critical component of next generation sequencing (NGS) library preparation workflows, as insufficient washing or cross contamination can adversely affect library quality and downstream sequencing results.
This technote describes the evaluation of the G.PURE for magnetic bead–based PCR cleanup, focusing on dispensing precision, washing performance, DNA recovery, and contamination control under conditions relevant to NGS library preparation.
Dispensing performance was independently verified, demonstrating high precision and accuracy across low volume dispensing ranges, enabling reliable automation of critical and high value reagents. Alternating wells containing amplified PCR products or no template controls were used to simulate contamination critical library cleanup conditions. Following automated bead cleanup and washing, residual DNA levels were quantified to assess washing efficiency and DNA carryover. Comparison to manual workflows demonstrated that automated bead cleanup using the G.PURE provides improved consistency and higher DNA recovery, highlighting the benefits of standardized liquid handling and reduced operator variability.
Overall, the results demonstrate high dispensing precision, efficient DNA cleanup, and supporting the suitability of the G.PURE for automated, bead based NGS library preparation workflows.
