Bead-based DNA cleanup has become a cornerstone of modern molecular biology workflows, offering reliable removal of primers, nucleotides, salts, and enzymes from nucleic acid preparations. Manual execution of these protocols, however, introduces variability that can compromise downstream results — particularly in sensitivity-demanding applications such as next-generation sequencing (NGS).
Here we describe how the G.PURE automated magnetic bead processing system, supported by the I.DOT Non-Contact Dispenser, enables consistent, scalable, and cost-effective bead-based cleanup across a wide range of applications and plate formats up to 384 wells. We present validated integration with five commercially available library preparation kits — including the Twist Enzymatic Fragmentation 2.0 Kit, Illumina DNA Prep, SMART-Seq HT, SMART-Seq HT PLUS, and NEBNext Ultra II — demonstrating broad workflow compatibility and end-to-end automation at up to 1/5 reaction volume in 384-well format.
